<p dir="ltr">This dataset accompanies the doctoral research thesis titled <i>“Elucidating genetic interactions in mycobacterial DNA metabolism using combinatorial CRISPR interference.”</i> <br><br><b>Study overview</b></p><p dir="ltr">A high-throughput inducible CRISPR interference (CRISPRi) screening approach was undertaken in the non-pathogenic mycobacterial model, <i>Mycobacterium smegmatis</i>, to map genetic interactions with the DNA repair complex called the mutasome. Whole-genome CRISPRi libraries were screened in six genetic backgrounds, including targeted gene-deletion mutants lacking <i>imuB</i> or <i>dnaE2</i> or inducible CRISPRi-mediated knockdown of <i>dnaE2. </i>DNA damage was induced by low-dose moxifloxacin (0.031 µg/mL), a fluoroquinolone used in the treatment of drug-resistant tuberculosis.</p><p dir="ltr"><b>Linked data:</b></p><p dir="ltr">sgRNA_info: sgRNA sequence and identifiers of the whole-genome CRISPRi library</p><p dir="ltr">raw_counts_all: Raw read counts obtained from Illumina sequencing files (FASTQ) of all repeats and conditions of the CRISPRi libraries processed in MAGeCK (version 0.5.9).</p><p dir="ltr">Source data of figures: Processes data underlying figures presented in the thesis, including normalized counts, interaction estimates, essentiality calls and significance metrics across experimental conditions.</p>
