This data was obtained from a an internal validation study for the Qubit™ 1X dsDNA HS Assay Kit as part of a molecular autopsy workflow. Therefore, control Lambda DNA, DNA extracted from forensic samples and NGS library samples were used for the validation experiments. This study was approved by the Faculty of Health Sciences Human Research Ethics Committee. Lambda DNA samples were diluted to various concentrations and extracted DNA and NGS library samples were diluted to fit within the assay's dynamic range. All samples were quantified using the Qubit™ 1X dsDNA HS Assay Kit on the Qubit™ 4 Fluorometer. The measured concentrations were used to determine the accuracy, intra- and inter-assay precision, range and sensitivity of the assay. The samples were also quantified using qPCR and TapeStation for comparative purposes.
